Tumilowicz, Joseph J. (Children's Hospital of Philadelphia, Philadelphia, Pa.), and Klaus Hummeler. Heterogeneity of apparently complete poliovirus particles. J. Bacteriol. 87:1105-1113. 1964.-A chromatographic procedure was developed for separating the N from the H complement-fixing antigen of poliovirus. This procedure concomitantly effected some separation of classes of N particle. The ratio of physical virus particles (PVP) to plaque-forming units (PFU) in N-reactive fractions varied from 6 to 51. Fractions with the lowest PVP-PFU ratios represented a considerable enrichment of PFU, when compared with the original ratio in each experiment. A direct relationship was found between the ratio of complement-fixing units of N antigen [CFU(N)] to 10(10) PVP and the ratio of PFU to 10(10) PVP for most of the N-reactive fractions. Large differences among the PVP-CFU(N) ratios, along with relatively constant PFU-CFU(N) ratios for most of the N fractions, indicated that N antigen is not distributed equally among non-H particles. Two possibilities, both compatible with the results, were discussed for the manner in which N antigen might be distributed. An absolute value of 10(7) PVP-CFU(N) was proposed for particles with a PVP-PFU ratio of 1.

Poliovirus Particles do not Form in Preinfected Cells with Reovirus
Background and Aims: Inhibition of viral growth in coinfected cells with two different viruses has been described. This phenomenon known as viral interference can occur in several virus host systems such as interference of enterovirus infection on poliovirus vaccine strains. In this study we superinfected reovirus infected HeLa cells with poliovirus to determine if poliovirus can replicate in s...
متن کاملDefective interfering particles of poliovirus. IV. Mechanisms of enrichment.
Infection of HeLa cells by mixtures of standard poliovirus and defective, interfering (DI) poliovirus particles leads to a higher ratio of DI particles in the progeny than in the inoculum. The extent of this enrichment could be varied by various manipulations of the co-infected cells. At any time during the infection cycle, virions made within short times after addition of radioactive uridine w...
متن کاملComparative poliovirus permeability of silver, polycarbonate, and cellulose membrane filters.
Three types of filters, silver, polycarbonate, and cellulose, were evaluated for permeability toward poliovirus suspended in water, salt-containing, and proteinaceous solvents. The ability of virus to pass through cellulose filters depended on the suspending medium; virus did not pass through cellulose filters in either water or salt solution, whereas the use of protein solutions increased filt...
متن کاملSynthesis of immunogenic, but non-infectious, poliovirus particles in insect cells by a baculovirus expression vector.
A baculovirus expression vector (AcLeon) derived from Autographa californica nuclear polyhedrosis virus (AcNPV) was prepared containing the complete 6.6 kb coding region of the P3/Leon/37 strain of poliovirus type 3 placed under the control of the AcNPV polyhedrin promoter. The recombinant virus was used to infect Spodoptera frugiperda insect cells. As demonstrated by use of the appropriate ant...
متن کاملMechanism of resistance to enteroviruses of some primate cells in tissue culture.
Two cell lines, M10-45-2 and L-41, were studied, each of which possessed specific resistance either to poliovirus or to coxsackievirus. Infection of M10-45-2 cells with poliovirus ribonucleic acid (RNA) and L-41 cells with infectious coxsackievirus RNA was accompanied by production of complete viruses in each of the resistant cell lines. During incubation of the cells with the virus to which th...
